Isolation and Identification of Pathogen of Ophiopogon japonicus Black Spot and Control Effect of Streptomyces diastatochramogenes

Abstract: Black spot disease is a common disease in the planting base of Ophiopogon japonicus in Cixi City,Zhejiang Province.It spreads rapidly and seriously affects the yield and quality of O.japonicus.In order to effectively control the disease,the infected tissue culture,pathogen isolation and purification,Koch's rule verification,the pathogen morphological observation and gene sequence analysis (ITS,TEF-1α,RPB2,Alt a 1,His 3,ATP) were carried out,and the pathogen of O.japonicus black spot was identified as Alternaria alternata.The metabolites of Streptomyces diastochromogenes 1628 obtained in previous study were used for O.japonicus black spot biocontrol.The results showed that S.diastochromogenes 1628 had a strong inhibitory effect on mycelial growth of A.alternata,with EC50 value of 0.764%(the ratio of volume).The corrected control efficacies on black spot of O.japonicus 7 and 14 days after treatment were 74.08% and 65.28%,respectively.The above results provide a basis for the subsequent prevention and control of O.japonicus black spot.
