Mechanism of Allelopathic Inhibition Effect of Tobacco (Nicotiana tabacum L.) Extracts on Phytophthora infestans

Abstract: In order to explore the inhibitory effect and mechanism of tobacco on Phytophthora infestans,the extracts of tobacco roots and leaves were used to treat P. infestans in each generation and cultured continuously for multiple generations,and the colony diameter and virulence of each generation were measured to screen the attenuated strain.The physiological changes and mycelium morphology of the attenuated strain were investigated under medium conditions,and the transcriptome was sequenced.The results showed that the colony diameter and virulence of P.infestans in each treatment were reduced under consecutive treatment by tobacco extracts for 8 generations.Among them,the disease index of the 7th and 8th generations of PI-R treatment was less than 10,so it was determined that the 8th generation of PI-R treatment was an attenuated strain PIAS-R of P. infestans.Compared with CK strain,the spore germination rate,biomass,and cell wall degrading enzymes activities of PIAS-R strain decreased significantly,and the mycelium morphology was obviously distorted.Transcriptomic analysis showed that 1086/1565 genes were significantly up/down-regulated in PIAS-R strain,respectively.Differentially expressed genes were enriched in 44 GO functional terms and 83 KEGG pathways.Tobacco could inhibit the physiological characteristics,destroy the mycelium morphological structure,and affect internal genes expression of P. infestans,which leads to the weakening of its virulence to the host.
