Effects of Temperature on Growth and Pathogenicity of Two Strains of Xenorhabdus bovenii

Abstract: To determine the influence of temperature on the growth and virulence of the symbiotic bacteria, as well as the optimal culture conditions, the experiment was conducted by plate and fermentation broth culture method under different temperature treatment conditions. Two strains of X. bovenii 0641TY and 0657L were cultured to study their growth rate and their hematotoxicity and gastrotoxicity to G. mellonella. The results showed that the average colony diameters of the X. bovenii 0641TY and 0657L with plate culture were 1653.34 and 834.84 μm respectively at 30 ℃. The two strains reached logarithmic growth stage at 6 and 12 h respectively at 30 ℃, and the growth rate was significantly higher than that at other temperatures.. The two strains cultured at 30 ℃ showed high gastrotoxicity to the larvae, and the corrected mortality rate reached 100% after 72 h of infection, while the hematotoxicity was weak, the mortality rate was 50% and 46.6% respectively after 96 h of infection.Therefore, 30 ℃ was beneficial to the growth and virulence of the two strains of X. boveni.
