Screening of Bt Soil Granule for White Grub Control

Abstract: To develop a new Bt formulation and improve its control efficiency against soil pest grub, the Bt185-gfp strain expressing the green fluorescent protein was constructed, and a special formulation for control of soil grub pest was developed using the peanut-meal that showed excellent multiplication for the Bt strain. There was no significant difference in Bt multiplication between 1% peanut-meal (W/V) and LB medium, indicating that the peanut-meal was a nutrient-rich and suitable material for Bt multiplication. When the peanut-meal and Bt spore powder were mixed together and suppressed into granules, the Bt in the granules multiplied by 30 times under laboratory sterile condition and by 40 times in soil. The granules caused a 61.11% mortality of the pest, which was much higher than the 19.44% mortality resulted from granules without peanut-meal, even when the two kinds of granules were of the same initial amount of Bt. We also found that the expanding process did not affect Bt multiplication, and produced Bt population size not different from that obtained from the sterilization process. In addition, the peanut-meal was also suitable for multiplication of other bio-control strains. This study provides a new way to develop a new formulation for soil pest and disease control.
