Identification of Lipopeptides of Endophytic Bacillus amyloliquefaciens BEB33 and Antifunal Activity against Fusarium oxysporum f. sp. cubense

Abstract: To investigate biocontrol mechanism of endophytic Bacillus amyloliquefaciens strain BEB33, the genes related to the synthesis of lipopeptides were detected and the lipopeptides were analyzed by matrix-assisted laser desorption ionization-time-of-flight mass spectrometry. The effects of crude lipopeptide extracts on the morphology of Fusarium oxysporum f. sp. cubense race4 (FOC4) and fusaric acid synthesis were investigated, and the biocontrol potential of crude extracts against banana wilt was also evaluated. The results showed that there 4 lipopeptide synthases (surfactin, fengycin, iturin and bacillomycin D) were detected in BEB33 genome. However, only three different lipopeptides (surfactin, fengycin and bacillomycin D) were detected in BEB33 lipopeptide crude extraction from M9 culture of BEB33. After treatment with BEB33 lipopeptide crude extraction, FOC4 hyphae became enlargement and could be dyed with trypan blue. The crude lipopeptide extract could reduce the yield of fusaric acid by 70%. The crude lipopeptide extract had obvious control effect on banana wilt. The disease incidence was reduced by 47.67%, 23.22% and 36.56%, respectively, and the disease index was decreased by 38.09, 28.56 and 35.87, respectively. These results provide a theoretical basis for further research and development of biocontrol agents for banana wilt disease.
