链霉菌FJ-10的鉴定及其对大豆炭疽病的防效

Identification of Streptomyces FJ-10 and Its Control Efficacy on Soybean Anthracnose

摘要: 为筛选对引起大豆炭疽病的不同炭疽菌具有抑菌活性的放线菌。采用土壤稀释涂布法和平板对峙法分离、筛选对大豆平头炭疽菌和胶孢炭疽菌具有抑制效果的放线菌菌株,利用表观分类和分子分类法对筛选的放线菌进行分类鉴定,同时测定候选菌株的抑菌谱和室内盆栽防效。研究结果显示,初筛获得15株对大豆炭疽病菌具有明显抑制作用的放线菌,其中菌株FJ-10的抑制作用最强,对大豆平头炭疽菌和胶孢炭疽菌的活体抑制率为86.25%和87.09%,无菌发酵液抑制率为82.93%和77.89%,菌株表观特征与灭癌素链霉菌Streptomyces gancidicus相似,16S rRNA和rpoB基因序列与S. gancidicus相应序列的相似度(同源性)分别为99.93%和99.76%,在系统发育进化树上与S. gancidicus属同一分支。最终将FJ-10鉴定为灭癌素链霉菌S. gancidicus。FJ-10对大豆炭疽病的盆栽防治效果为70.32%。FJ-10抑菌谱广,除了对大豆炭疽病具有防治效果外,还对多种植物病原菌均具有良好的抑制作用,具有良好的推广应用前景。

Abstract: o screen actinomycetes strains against diverse Colletotrichum species that cause soybean anthracnose, the soil dilution spread plate method and the plate confrontation method were used to isolate and screen actinomycete strains with inhibitory effects on C. truncatum and C. gloeosporioides. The selected actinomycetes were classified and identified by phenotypic classification and molecular classification. Simultaneously, the antifungal spectrum of the candidate strains and their control efficacy in indoor potted plants were determined. The results indicated that 15 actinomycete strains with distinct inhibitory effects on soybean anthracnose pathogens were initially isolated. Among these, strain FJ-10 demonstrated the strongest inhibitory activity, with the in-vivo inhibition rates of 86.25% and 87.09% against C. truncatum and C. gloeosporioides, respectively, while those of its sterile fermentation broth were 82.93% and 77.89%, respectively. The apparent characteristics of the strain were similar to those of Streptomyces gancidicus. The similarities (homology) between the 16S rRNA and rpoB gene sequences of strain FJ-10 and those of S. gancidicus were 99.93% and 99.76%, respectively. On the phylogenetic tree, strain FJ-10 was grouped within the same branch as S. gancidicus. Consequently, FJ-10 was identified as S. gancidicus. In potted plant experiments, FJ-10 exhibited a 70.32% control efficacy against soybean anthracnose. FJ-10 possessed a broad antifungal spectrum, which had significant inhibitory effects on a wide range of plant pathogens. Given these findings, FJ-10 holds great promise for development and application in the field.
