Identification of anti-magnaporthe oryzae bacteria of Bacillus amyloliquefaciens from traditional Chinese medicine and Characterization of its Active components

Abstract: In order to look for new antagonistic microbial resources against rice blast, a strain CWJ2 with good performance was isolated from traditional Chinese medicine extracts. Dural culture test showed strain CWJ2 could inhibit the mycelium growth of Magnaporthe oryzae with the rate of 69.51%. The strain was identified as Bacillus amyloliquefaciens by colony and strain morphology, 16s rDNA and gyrB gene sequence. Biological activity tests showed that the germ-free fermentation filtrate of strain CWJ2 could significantly inhibit the mycelium growth and the spores germination of M. oryzae. 10% fermentation filtrate of the strain could inhibit the mycelium growth with the rate of 100%, and 2% fermentation filtrate could lead to a significant reduction in mycelial branching, and shorter, enlarged or deformed mycelium. Under high temperature, proteinase K, ultraviolet, acid and alkali treatments, the antagonistic activity of the fermentation filtrate of CWJ2 strain did not change significantly. All the results suggested that the strain CWJ2 had good inhibition effect against M. oryzae and its antagonistic components were very stable, which was expected to be developed as a biocontrol agent for rice blast control.
